A Reverse-Transcription Loop-Mediated Isothermal Amplification Technique to Detect Tomato Mottle Mosaic Virus, an Emerging Tobamovirus

Tomato mottle mosaic virus (ToMMV) is an emerging seed-transmissible tobamovirus that infects tomato and pepper. Since the first report in 2013 Mexico, ToMMV has spread worldwide, posing a serious threat to production of both crops. To prevent this virus, early accurate detection infection required. In study, we developed method for based on reverse-transcription loop-mediated isothermal amplification (RT-LAMP). A LAMP primer set was designed target genomic region spanning movement protein coat genes, which highly conserved sequence unique ToMMV. This RT-LAMP achieved 10-fold higher sensitivity than conventional RT-polymerase chain reaction methods obtained high specificity without false positives closely related tobamoviruses or healthy plants. can detect within 30 min direct sampling infected leaf using toothpick therefore does not require RNA purification. Given its sensitivity, specificity, simplicity, rapidity, study expected be valuable point-of-care testing field surveys large-scale testing.
